I'm running Vista 64 right now and my old Colorvision Spyder isn't supported but I still have XP loaded so I can dual boot if needed. If I calibrate and create the profile in XP, can I use that same profile on the Vista side and it be accurate?

I would run it in Vista then do a screen print of both opened with PS and your image viewer of choice and see if there is any difference.
It might work and it will prob be better than no profile until you upgrade your calibration tools.
